Description

Assay Genie's PKC zeta (Phospho-Thr560) Colorimetric Cell-Based ELISA Kit is specifically designed for the accurate detection of phosphorylated PKC-zeta (Thr560) levels in cell lysates and tissue lysates, with high sensitivity and specificity. The cell-based, lysate-free assay uses an indirect ELISA format, in which the target is captured by a phospho-specific primary antibody and detected via an HRP-conjugated secondary antibody that catalyses a colorimetric reaction on substrate addition. It can be used to measure the relative amounts of phosphorylated PKC zeta in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on PKC zeta phosphorylation.

The kit is reactive with Human, Mouse, and Rat samples and uses a colorimetric detection method read at 450 nm, with a dynamic range of > 5000 Cells. Adherent cells are supported as the sample type. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalised to cell amounts to correct for plating differences.

PKC-zeta is an AGC kinase of the protein kinase C family and an atypical PKC whose activity is not regulated by Ca2+, PS, DAG, or phorbol esters, and it is constitutively active. Phosphorylation at Thr560 plays a crucial role in regulating its activity and function, and dysregulation of PKC-zeta phosphorylation has been implicated in cancer, diabetes, and inflammatory disorders. The target gene symbol is PRKCZ, the sub type is Phospho, and the research area is immunology.
