PKC zeta Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit enables qualitative detection of protein kinase C zeta (PKC zeta) directly in cultured adherent cells without the need for prior lysate preparation. The assay uses an indirect ELISA format in which PKC zeta is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies; an HRP-catalysed colorimetric reaction, read at 450 nm, provides the readout. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be applied after the substrate step to assess cell density and adjust for plating variation.

Reactivity is reported for human, mouse and rat samples, with a stated dynamic range of > 5000 cells per well. It is supplied in a 96-well microplate format and is stored at 4 °C with a stated stability of 6 months. PKC zeta is described as an atypical, calcium- and diacylglycerol-independent member of the protein kinase C family of serine/threonine kinases and is implicated in cellular proliferation, differentiation and apoptosis, with dysregulation linked to cancer, diabetes and autoimmune disorders. The kit is therefore suited to signal transduction studies and to screening the effects of various treatments, inhibitors (such as siRNA or chemical compounds) or activators on PKC zeta.
