PKC theta (Phospho-Thr538)Fluorometric Cell-Based ELISA Kit

This phospho-specific cell-based assay from Assay Genie is designed to qualitatively measure Phospho-PKC θ (Thr538) levels in cultured cells using a fluorometric, indirect ELISA format. The kit is a lysate-free, high-throughput and sensitive solution for monitoring PKC theta phosphorylation and expression profiles, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on PRKCQ phosphorylation.

PKC theta (Phospho-Thr538) is captured by target-specific primary antibodies, while Dye 1- and Dye 2-conjugated secondary antibodies bind the Fc region of the primary antibody to enable fluorometric detection. For data normalization, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control, as well as an antibody against the non-phosphorylated counterpart of PKC theta (Phospho-Thr538). Supplied components include a 96-well black cell culture clear-bottom microplate, 10X TBS, Quenching Buffer, Blocking Buffer, 15X Wash Buffer, Primary Antibody Diluent, 100x Anti-Phospho Target Antibody, 100x Anti-Target Antibody, Anti-GAPDH Antibody, Dye-1 Conjugated Anti-Rabbit IgG Antibody, Dye-2 Conjugated Anti-Mouse IgG Antibody, and Adhesive Plate Seals.

Reactivity has been validated for Human, Mouse and Rat samples, and the assay is read on a fluorescent plate reader at Ex/Em 651/667 and 495/521. The dynamic range is > 5000 Cells, and recommended storage is 4°C for 6 months. The target gene PRKCQ corresponds to Gene ID 5588 and UniProt ID Q04759.
