Description

Assay Genie offers a cell-based ELISA kit for the detection and quantification of phosphorylated PKC theta at serine 676. The kit is lysate-free and high-throughput, using an indirect ELISA format in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction. It is intended for adherent cells and reacts with human, mouse, and rat samples.

The assay has a dynamic range of > 5000 cells and uses colorimetric detection at 450 nm in a microplate format. A monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density for normalization of absorbance values to cell amounts. The kit supports measurement of relative levels of phosphorylated PKC theta in cultured cells and screening of treatments, inhibitors (such as siRNA or chemicals), or activators for their effects on PKC theta phosphorylation.

PKC theta, encoded by the PRKCQ gene (Gene ID: 5588, UniProt: Q04759), is a calcium-independent, phospholipid-dependent serine/threonine kinase of the PKC family. It plays a role in T-cell receptor signaling and T-cell activation, including activation of the transcription factors NF-kappaB and AP-1. Phosphorylated PKC theta at serine 676 is involved in immune responses and inflammatory processes, and its dysregulation has been linked to autoimmune disorders and cancer.
