Description

A colorimetric cell-based ELISA from Assay Genie is designed for the detection and quantification of PKC epsilon phosphorylated at serine 729 in cell lysates. The assay uses an indirect ELISA format, in which phospho-specific and total primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. A Crystal Violet whole-cell staining step allows normalization of absorbance values to cell density, and a monoclonal anti-GAPDH antibody is included as an internal positive control.

The procedure is lysate-free and high-throughput, supporting measurement of relative phosphorylated PKC epsilon levels in cultured cells and screening of the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on PKC epsilon phosphorylation. Reactivity is reported for human, mouse, and rat, with a stated dynamic range of > 5000 cells. The kit is intended for use with adherent cells and supports cell biology research into PKC epsilon signaling, a pathway implicated in processes such as cell proliferation, differentiation, and apoptosis and in conditions including cancer, cardiovascular disease, and neurological disorders.
