Description

From Assay Genie, this Cell-Based ELISA Kit provides a colorimetric approach for detecting and quantifying PKC epsilon protein levels, supporting research into cellular signalling and the role of this isoform in disease biology. PKC epsilon is a calcium-independent, phospholipid-dependent AGC kinase of the PKC family that regulates cell proliferation, differentiation, and apoptosis.

Reactive with Human, Mouse, and Rat samples and intended for use with adherent cells, the assay detects PKC epsilon at 450 nm in an indirect ELISA format. PKC epsilon is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyse a colour reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. The dynamic range is > 5000 Cells.

The kit uses a 96-Well Microplate format and is rated for storage at 4°C for 6 months. Accompanying background notes indicate that PKC epsilon is activated by inflammatory mediators and involved in nociceptive functions, with PKC-epsilon null mice displaying decreased hypoxic pulmonary vasoconstriction. The target is encoded by PRKCE (Gene ID: 5581; UniProt ID: Q02156), located at chromosomal position 2p21 with a molecular weight of 83,674 Da.
