Description

A cell-based ELISA from Assay Genie is designed for the accurate quantification of PKC delta phosphorylation at Ser645 in human cells. The assay uses an indirect ELISA format in which phospho-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Detection is by colorimetric measurement at 450 nm with a dynamic range of > 5000 cells.

The kit is described as lysate-free, high throughput, and sensitive, and is used with adherent cells. It allows measurement of the relative amounts of phosphorylated PKC delta in cultured cells and can be applied to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), and activators on PKC delta phosphorylation. For data normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining step can be used to determine cell density after the colorimetric measurement.

PKC delta is a key protein kinase C isoform involved in cell growth, differentiation, and apoptosis, and phosphorylation at Ser645 is a critical regulatory event that modulates its activity and function. Dysregulation of PKC delta phosphorylation has been implicated in cancer, inflammation, and neurological disorders, making it relevant as a potential therapeutic target and biomarker for disease progression. The target corresponds to gene symbol PRKCD (Gene ID: 5580; UniProt ID: Q05655).
