Description

An in-cell ELISA kit from Assay Genie provides qualitative detection of protein kinase C alpha (PKC alpha) expression in cultured cells, with no lysate preparation required. The assay uses an indirect ELISA format: PKC alpha-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction measured at 450 nm. The kit reacts with human, mouse, and rat samples and has a dynamic range of > 5000 cells. A monoclonal anti-GAPDH antibody serves as an internal positive control for normalization, while Crystal Violet whole-cell staining is used to determine cell density. The kit is suited to measuring relative PKC alpha amounts in cultured cells and to screening the effects of treatments, inhibitors (including siRNA or chemicals), or activators. Storage is at 4°C for 6 months, and the format is a 96-well microplate.

PKC alpha is a key enzyme regulating cell growth, proliferation, and differentiation, and its dysregulation has been implicated in cancer, diabetes, and neurodegenerative disorders. The protein is encoded by the PRKCA gene (Gene ID: 5578; UniProt ID: P17252), with a reported molecular weight of 76,750 Da. It is a classical, calcium-dependent AGC kinase of the PKC family, activated by phosphatidylserine, diacylglycerol, and phorbol esters, and contains a pseudo-substrate autoinhibitory domain that binds the catalytic domain to prevent activation in the absence of cofactors or activators.
