Description

A colorimetric cell-based ELISA kit from Assay Genie is designed for the accurate detection of phosphorylated PKA-R2beta (Ser113) levels in cell lysates. The kit targets PKA-R2beta (Phospho-Ser113) and is reactive with Human, Mouse, and Rat samples, and is suitable for adherent cells, making it a valuable tool for researchers studying protein kinase A (PKA) signaling pathways.

Detection uses an indirect ELISA format, in which PKA-R2beta (Phospho-Ser113) is captured by target-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Through this binding, the HRP enzyme catalyzes a colorimetric reaction upon substrate addition, with signal read at 450 nm. The assay has a dynamic range of > 5000 Cells, is lysate-free and high throughput, and can be used to measure relative amounts of phosphorylated PKA-R2beta in cultured cells as well as to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PKA-R2beta phosphorylation.

For normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and the Crystal Violet whole-cell staining method may be used to determine cell density. The target, PRKAR2B (Gene ID: 5577; UniProt ID: P31323), encodes the cAMP-dependent protein kinase type II-beta regulatory subunit with a molecular weight of 46kDa, a regulatory subunit of cAMP-regulated protein kinase involved in cellular processes including cell growth, metabolism, and gene expression.
