PKA alpha/beta CAT (Phospho-Thr197)Colorimetric Cell-Based ELISA Kit

This Assay Genie cell-based ELISA kit is designed for the detection of phosphorylated PKA alpha/beta CAT (Phospho-Thr197) in cell lysates and tissue samples. Using a colorimetric indirect format, the assay captures the phospho-target with a phospho-specific primary antibody, while an HRP-conjugated secondary antibody bound to the Fc region of the primary antibody catalyses a colorimetric reaction upon substrate addition. This enables qualitative measurement of PKA alpha/beta CAT phosphorylation and expression in cells.

The kit reacts with human, mouse and rat samples and is intended for use with adherent cells. It has a dynamic range of > 5000 cells and detection is read at 450 nm. It can be used to measure relative amounts of phosphorylated PKA alpha/beta CAT in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PKA alpha/beta CAT phosphorylation.

An anti-GAPDH monoclonal antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be normalised to cell counts.
