Description

A cell-based ELISA kit from Assay Genie designed for the qualitative measurement of PIAS3 (Protein Inhibitor of Activated STAT 3) protein expression in cultured cells. The kit employs an indirect ELISA format in which PIAS3 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. It is intended for adherent cell samples and can be used to assess the relative amounts of PIAS3 in cell lysates as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on PIAS3 expression.

The assay is reactive with human, mouse and rat samples and has a stated dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density and further adjust for plating differences. The kit is supplied in a 96-well microplate format, is stored at 4°C with a stated stability of 6 months, and is positioned for research in the area of epigenetics and nuclear signaling.

PIAS3 is a member of the PIAS family of transcriptional modulators and functions as an E3-type small ubiquitin-like modifier (SUMO) ligase, playing a critical role in the regulation of STAT3 signaling by inhibiting STAT3 DNA-binding activity. It is therefore of relevance to studies of cell growth, differentiation and apoptosis, as well as to research on diseases such as cancer and inflammatory disorders.
