Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the accurate measurement of PIAS1 levels in cell lysates and culture supernatants. The assay targets PIAS1, also known as protein inhibitor of activated STAT 1, a critical regulator of the JAK/STAT signaling pathway that is involved in immune response, apoptosis, and cell proliferation. Aberrant PIAS1 expression has been linked to diseases including cancer and inflammatory disorders. The kit is reactive with human and mouse samples, uses a colorimetric detection method at 450 nm, supports a dynamic range of > 5000 cells, and is supplied in a 96-well microplate format intended for storage at 4°C with 6 months of stability. It is intended for use with adherent cells.

The kit provides a lysate-free, high-throughput, and sensitive workflow for detecting PIAS1 protein expression profiles in cultured cells. It can be used to measure relative amounts of PIAS1 and to screen for the effects of treatments, inhibitors such as siRNA or chemicals, or activators. Detection proceeds by an indirect ELISA format, in which PIAS1-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. An anti-GAPDH monoclonal antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell amounts.

The kit falls within the Epigenetics and Nuclear Signaling research area. Associated database identifiers for PIAS1 include Gene ID 8554 and UniProt ID O75925.
