Description

A colorimetric, cell-based ELISA kit from Assay Genie designed for the qualitative measurement of phosphorylated GAB1 at tyrosine 659 in cultured adherent cells. The kit provides a lysate-free, high-throughput indirect ELISA workflow in which target phosphorylation is captured by a phospho-specific primary antibody and detected using an HRP-conjugated secondary antibody that catalyses a colorimetric reaction at 450 nm.

GAB1 is a key adaptor protein involved in intracellular signalling cascades downstream of receptor-type kinases, with phosphorylation at Tyr659 being particularly important in cell proliferation and survival pathways. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. The kit is reactive with human, mouse and rat samples and has a stated dynamic range of > 5000 cells. It is supplied in a microplate format with primary antibody diluent, wash buffer, blocking buffer, quenching buffer, substrate, stop solution and adhesive plate seals included.

Suitable applications include measuring relative levels of phosphorylated GAB1 in cultured cells and screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on GAB1 phosphorylation. The associated GAB1 gene is recorded under Gene ID 2549 with UniProt accession Q13480 and a molecular weight of 80,005 Da.
