Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is a lysate-free, high throughput assay designed for the rapid detection of PGBD1 levels in cell lysates and culture supernatants. The kit offers high sensitivity and specificity for measuring relative PGBD1 protein expression in cultured cells, and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PGBD1 expression.

Qualitative determination is achieved through an indirect ELISA format in which PGBD1 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Upon substrate addition, the conjugated HRP catalyses a colorimetric reaction read at 450 nm. A monoclonal anti-human GAPDH antibody is supplied as an internal positive control for normalization of target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density for further adjustment of plating differences.

The kit is configured for adherent cells, with reactivity against human samples and a stated dynamic range of > 5000 cells. Storage is at 4 °C for 6 months. PGBD1 (piggyBac transposable element-derived protein 1) is a member of the piggyBac transposase family involved in DNA transposition and gene regulation, and is specifically expressed in the brain, with reported links to diseases such as cancer and genetic disorders.
