PFKFB1/4 Colorimetric Cell-Based ELISA

A cell-based immunoassay from Assay Genie enables detection of PFKFB1/4 protein expression in cultured cells using a Colorimetric 450 nm read-out. The kit is a lysate-free, high-throughput and sensitive format for measuring the relative amounts of PFKFB1/4 in cultured cells and for assessing the effects that various treatments, inhibitors or activators have on the target. Qualitative determination of PFKFB1/4 concentration is achieved via an indirect ELISA format, in which target-specific primary antibodies capture PFKFB1/4 and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody.

Reactivity covers Human, Mouse and Rat, and the assay is designed for adherent cells. The dynamic range is > 5000 Cells. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising target absorbance values, and following colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density. After staining, absorbance values can be normalised to cell amounts to adjust for plating differences.

PFKFB1 is a key enzyme involved in glycolysis that plays a role in regulating glucose metabolism and cell proliferation, and its dysregulation has been linked to cancer, diabetes and cardiovascular disorders. The kit is presented as a tool for studying the role of PFKFB1 in these conditions and exploring potential therapeutic interventions. Storage and stability are 4°C for 6 Months.
