Description

Assay Genie provides a Cell-Based ELISA kit intended for the qualitative detection of PDLIM1 in cell lysates, with a workflow that does not require separate lysate preparation. The assay uses an indirect ELISA format: a PDLIM1-specific primary antibody captures the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric signal at 450 nm upon substrate addition. The kit is validated for Human, Mouse, and Rat reactivity and is run on a 96-well microplate format.

The dynamic range is greater than 5000 cells, and the kit is intended for measuring relative PDLIM1 expression in cultured cells as well as screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. Because the assay is qualitative, two approaches are provided for normalization: a monoclonal anti-human GAPDH antibody serves as an internal positive control for target absorbance values, and a Crystal Violet whole-cell staining step is used to determine cell density, allowing absorbance values to be normalized to cell amount and adjusting for plating differences.

PDLIM1, also referred to as CLP36 or CLIM1, is a PDZ and LIM domain protein involved in cell signaling and actin cytoskeleton organization, with dysregulation implicated in cancer and cardiovascular disorders. The kit is stored at 4 °C with a stated stability of 6 months, and the expected sample type is adherent cells, supporting studies of PDLIM1 function and its role in disease pathways.
