PDK1 Colorimetric Cell-Based ELISA Kit

A colorimetric cell-based ELISA kit from Assay Genie designed to detect PDK1 levels in cell lysates. PDK1 is described as a serine/threonine kinase that acts as a master kinase, regulating cell growth, proliferation, and survival by phosphorylating and activating a subgroup of the AGC family of protein kinases, with targets including PKB/AKT, p70 ribosomal protein S6 kinase, and p90 ribosomal protein S6 kinase. The kit uses an indirect ELISA format in which PDK1 is captured by specific primary antibodies and detected via HRP-conjugated secondary antibodies, producing a colorimetric reaction read at 450 nm.

The assay is described as lysate-free and high-throughput, suitable for measuring the relative amounts of PDK1 in cultured cells as well as screening the effects of treatments, inhibitors, or activators on PDK1 expression. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control, and a Crystal Violet whole-cell staining step may be used to normalise absorbance values to cell density.
