Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the detection of phosphorylated PDGFRbeta at tyrosine 740 in cell lysates or tissue samples. It targets PDGFRbeta (Phospho-Tyr740) and shows reactivity with Human, Mouse, and Rat samples, making it suitable for a range of research applications. The assay uses a Colorimetric detection method measured at 450 nm with a dynamic range of > 5000 Cells and is intended for use with Adherent Cells.

The kit provides a lysate-free, high-throughput, Cell-Based ELISA format that detects PDGFRbeta protein phosphorylation and expression profiles in cultured cells. It can be used to measure the relative amounts of phosphorylated PDGFRbeta in cultured cells and to screen for the effects that various treatments, inhibitors such as siRNA or chemicals, or activators have on PDGFRbeta phosphorylation. Qualitative determination of PDGFRbeta (Phospho-Tyr740) concentration is achieved by an indirect ELISA format, in which PDGFRbeta (Phospho-Tyr740) is captured by specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values. Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density, enabling absorbance values to be normalised to cell amounts.
