Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the colorimetric detection of PDGFRbeta (Platelet-derived Growth Factor Receptor beta) in cultured cells. The assay is intended for adherent cell samples and provides a lysate-free, high-throughput workflow for measuring relative PDGFRbeta protein expression in cells, as well as for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on PDGFRbeta.

Detection is performed at 450 nm, with a stated dynamic range of > 5000 Cells. The assay uses an indirect ELISA format in which PDGFRbeta-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. Because the assay is qualitative, two normalisation approaches are provided: a monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density and adjust for plating differences. The kit is reactive with Human, Mouse, and Rat samples and is stored at 4°C for 6 months.

PDGFRbeta is a receptor tyrosine kinase of the PDGFR family that binds members of the platelet-derived growth factor family; ligand binding drives receptor dimerisation and autophosphorylation, regulating cell growth, actin reorganisation, migration, and differentiation. Associated database entries include Gene ID 5159 and UniProt ID P09619.
