Description

A high-quality cell-based ELISA from Assay Genie intended for the quantitative determination of PDGFRalpha phosphorylated at Tyrosine 713 in cell samples. The kit targets PDGFRalpha (Phospho-Tyr713) with human and mouse reactivity, and is intended for use with adherent cells. Detection is by colorimetric method measured at 450 nm, with a stated dynamic range of > 5000 cells.

The assay uses an indirect ELISA format and is described as a convenient, lysate-free, high-throughput and sensitive kit capable of detecting PDGFRalpha protein phosphorylation and expression profiles in cells. It can be used to measure relative amounts of phosphorylated PDGFRalpha in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PDGFRalpha phosphorylation. PDGFRalpha (Phospho-Tyr713) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing HRP to catalyze a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing target absorbance values. Following the colorimetric measurement of HRP activity, a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts. Database references for the target include Gene ID: 5175, UniProt ID: P16234, and OMIM: 173445, with gene symbol PDGFRA and sub-type Phospho.
