Description

A colorimetric cell-based (in-cell) ELISA from Assay Genie that detects PDGFRa protein phosphorylation at Tyrosine 849 as well as the expression profile of PDGFRa in cultured adherent cells. The kit operates in a lysate-free, high-throughput, sensitive format and uses an indirect ELISA approach: PDGFRa (Phospho-Tyr849) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction at 450 nm upon substrate addition. The dynamic range is > 5000 Cells, and the kit is reactive with Human, Mouse, and Rat samples.

The kit supports qualitative measurement of relative PDGFRa phosphorylation levels in cultured cells, making it suitable for screening the effects of various treatments, inhibitors (e.g., siRNA or chemicals), and activators on PDGFRa phosphorylation. Two normalization strategies are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step enables normalization of absorbance values to cell density.

PDGFRA is a receptor tyrosine kinase of the PDGFR family, with roles in cell growth, survival, and differentiation. Phosphorylation at Tyr849 is implicated in signaling pathways that regulate cell proliferation and migration, making the target relevant to research in areas such as cancer and fibrosis, as well as to broader drug development and therapeutic intervention efforts. Gene ID: 5156; UniProt ID: P16234; OMIM: 173490/607685; Unigene: Hs.74615.
