Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the detection of phosphorylated PDCD4 at serine 67 in cell lysates, with the assay optimised for use on adherent cells. The kit targets PDCD4 (Phospho-Ser67) and is reactive with human, mouse and rat samples. The manufacturer lists the research area as Cell Death.

Detection is by colorimetric measurement at 450 nm, with a stated dynamic range of > 5000 cells. The assay uses an indirect ELISA format in which PDCD4 (Phospho-Ser67) is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition. Because the Cell-Based ELISA is qualitative, two normalisation methods are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control, and a Crystal Violet whole-cell staining step is used to determine cell density and adjust for plating differences.

According to the database information supplied with the kit, PDCD4 (Programmed cell death protein 4) is a tumour suppressor localised to the nucleus in proliferating cells. The protein directly interacts with the RNA helicase eIF4A and inhibits assembly of the cap-dependent translation initiation complex, and dysregulation of PDCD4 phosphorylation has been implicated in cancer progression and drug resistance.
