Description

This cell-based ELISA kit from Assay Genie is designed for the accurate detection of phosphorylated PDCD4 (Ser457) levels in cell lysates and tissue homogenates, with reactivity against human, mouse, and rat samples. The assay uses a colorimetric detection method measured at 450 nm in an indirect ELISA format, in which the target is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The stated dynamic range is "> 5000 Cells".

Phosphorylation of PDCD4 at Ser457 is known to regulate its interactions with other proteins and its function in processes such as cell growth, differentiation, and apoptosis. Abnormal phosphorylation of PDCD4 has been implicated in various diseases, including cancer, inflammatory disorders, and autoimmune diseases, making the target relevant to the research area of cell death.

The kit includes a monoclonal antibody specific for human GAPDH as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density. The assay is intended for use with adherent cells. PDCD4 is annotated as a tumor suppressor protein (UniProt ID: Q53EL6, Gene ID: 27250) with a reported molecular weight of 50,576 Da.
