Description

This cell-based ELISA kit from Assay Genie is designed for the quantitative measurement of PBK/TOPK protein levels in cultured human cells. Using a Colorimetric detection method at 450 nm, the kit operates in an indirect ELISA format in which PBK/TOPK is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies, with a stated dynamic range of > 5000 Cells.

The assay is lysate-free and intended for use with adherent cells, allowing researchers to determine the relative amounts of PBK/TOPK in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on the target. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values may be normalized to cell amount, correcting for plating differences.

PBK/TOPK (PDZ-binding kinase, also known as TOPK) is a serine/threonine protein kinase implicated in cell proliferation, cell cycle regulation and tumorigenesis, and is reported to be active primarily during mitosis, where it phosphorylates MAP kinase p38. The kit is stored at 4°C with a stated stability of 6 Months, and the human target corresponds to Gene ID: 55872 and UniProt ID: Q96KB5, located at chromosome 8p21.2, with a reported molecular weight of 37,211 Da.
