Description

From Assay Genie, this cell-based ELISA kit enables qualitative measurement of paxillin protein expression in cultured cells, including adherent cell samples. Using an indirect ELISA format, the assay captures paxillin with target-specific primary antibodies and detects it through HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. It is suitable for use with cell lysates, tissue lysates, and cell culture supernatants, and can be used to screen the effects of treatments, inhibitors such as siRNA or chemicals, and activators on paxillin levels.

The kit is reactive with human, mouse, and rat samples, with detection read at 450 nm. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining method is provided to determine cell density. The microplate format has a dynamic range of > 5000 cells, and the kit is stable at 4 °C for 6 months.

Paxillin (PXN) is a multi-domain cytoskeletal protein that localizes primarily to focal adhesion sites, where it is phosphorylated by focal adhesion kinase (FAK) and participates in integrin signaling. The protein has a reported molecular weight of 68 kDa.
