The Assay Genie Pax-8 Transcription Factor Activity Assay quantifies the DNA-binding activity of the Pax-8 protein, a paired-box transcription factor essential for thyroid follicular cell differentiation, kidney organogenesis, and Müllerian duct development. This non-radioactive DNA-binding ELISA captures active Pax-8 from nuclear and cell lysates using an immobilized double-stranded oligonucleotide probe bearing the canonical Pax-8 paired-domain recognition sequence. Bound transcription factor is detected with an HRP-conjugated secondary antibody and a chromogenic substrate, producing a measurable absorbance change in each reaction.

Researchers use this kit to investigate transcriptional regulation in thyroid carcinoma, ovarian surface epithelial tumors, and renal cell carcinoma, where Pax-8 serves as both a lineage marker and a candidate driver of tumor cell proliferation. The format accommodates up to 96 Assays per kit with 100 µL of sample loaded per well, and includes pre-coated strip wells, nuclear extraction reagents, binding buffer, and standard controls. For best results, prepare nuclear extracts freshly and store all kit components at 4 °C; protect the substrate solution from light and avoid repeated freeze-thaw cycles of the standard and antibody reagents.
