Description

From Assay Genie comes a Cell-Based ELISA Kit designed for the precise quantification of Parkin levels in cell lysates and tissues. Parkin is a key protein involved in the regulation of mitochondrial quality control and the removal of damaged organelles through mitophagy. Dysregulation of parkin has been linked to neurodegenerative diseases, including Parkinson's disease, making it a critical biomarker for research into these conditions and potential therapeutic interventions.

This colorimetric assay operates at 450 nm and provides a convenient, lysate-free, high-throughput and sensitive workflow. It can be used to measure relative amounts of Parkin in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Parkin. Qualitative determination is achieved via an indirect ELISA format, in which Parkin-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the HRP enzyme to catalyze a colorimetric reaction upon substrate addition.

Reactivity is for human samples and the dynamic range is > 5000 Cells. Storage is at 4°C with 6 months stability. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, enabling absorbance values to be normalized to cell amounts. Associated database identifiers include Gene ID: 5071, UniProt ID: O60260, OMIM: 602544, and Unigene: Hs.132954. The kit is intended for use with adherent cells.
