PAK3 Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit is designed for the quantification of PAK3 protein levels in cultured cells using a colorimetric detection method read at 450 nm. The assay is reactive with human, mouse and rat samples and offers a dynamic range of > 5000 Cells. It is supplied in a 96-well microplate format with a storage stability of 4°C for 6 months, and is suited to adherent cell samples.

The kit employs a lysate-free, indirect ELISA format in which PAK3-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density for further normalisation. The qualitative assay can be applied to measure relative PAK3 amounts in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators.

PAK3 is a serine/threonine protein kinase of the STE20 family, acting as a key regulator of cytoskeletal dynamics, cell migration, adhesion and proliferation, and has been implicated in cancer, neurological and immune-related disorders.
