Description

Assay Genie offers a colorimetric, cell-based ELISA kit designed for the detection of phosphorylated PAK2 (p21-activated kinase 2) at Ser141 in cell lysates and tissue homogenates. The assay is described as highly sensitive and specific, and is intended for studying the role of PAK2 phosphorylation in cellular processes such as cell growth, survival, and cytoskeletal remodeling. Phosphorylation of PAK2 at Ser141 is reported to enhance its kinase activity and has been implicated in cancer progression, neurodegeneration, and immune response modulation.

The kit uses an indirect ELISA format and is lysate-free, allowing the relative amounts of phosphorylated PAK2 to be measured in cultured cells. It can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PAK2 phosphorylation. Detection is achieved colorimetrically at 450 nm via HRP-conjugated secondary antibodies. A monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density for normalization. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of "> 5000 Cells", and is suitable for adherent cell samples.

PAK2 is a protein kinase of the STE20 family, acting as a critical effector linking RhoGTPases to cytoskeleton reorganization and nuclear signaling, and is activated by proteolytic cleavage during caspase-mediated apoptosis. Associated database identifiers include Gene ID: 5062, UniProt ID: Q13177, OMIM: 605022, and Unigene: Hs.518530.
