PAK1 (Phospho-Ser204)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit enables qualitative detection of PAK1 phosphorylated at serine 204 in cultured cells without requiring lysate preparation. The assay uses a colorimetric readout at 450 nm and is suitable for adherent cell samples with a dynamic range of "> 5000 Cells". Reactivity has been validated for Human, Mouse, and Rat samples.

The indirect ELISA format employs primary antibodies specific for the phospho-Ser204 target, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyse a colour-producing reaction upon substrate addition. A monoclonal anti-GAPDH antibody is supplied as an internal positive control for normalising target absorbance, and a Crystal Violet whole-cell staining step is included to determine cell density so that absorbance values can be normalised to cell amounts.

Phosphorylation of PAK1 at serine 204 modulates its kinase activity, and the kit supports investigation of PAK1-mediated signalling in cancer, neurodegenerative disorders, and other conditions where dysregulation of this pathway is implicated. It is suited to screening the effects of various treatments, siRNA or chemical inhibitors, or activators on PAK1 phosphorylation in cultured cells.
