Description

A Cell-Based ELISA Kit from Assay Genie designed for studying the phosphorylation status of p95-NBS1 at Ser278 in cellular samples. p95-NBS1 plays a crucial role in the DNA damage response pathway and the repair of DNA double-strand breaks, with phosphorylation at Ser278 known to regulate its function in DNA damage repair. The kit supports research into cancer biology, genomic instability, and radiation therapy.

The assay is a cell-based, colorimetric ELISA with reactivity for human samples. It has a dynamic range of > 5000 Cells and uses colorimetric detection at 450 nm, supplied in a 2 x 96-Well Microplates format. The kit is suitable for adherent cells and operates as a lysate-free, high-throughput, sensitive assay. It detects p95-NBS1 protein phosphorylation and expression profile in cells, and can be used to measure the relative amounts of phosphorylated p95-NBS1 in cultured cells, as well as to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on p95-NBS1 phosphorylation.

Qualitative determination is achieved by an indirect ELISA format, in which the target is captured by specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density for further normalization. The target corresponds to the NBN gene (Gene ID: 4683; UniProt ID: O60934), with a sub-type classification of Phospho.
