Description

A cell-based colorimetric ELISA kit from Assay Genie for the qualitative detection of p90 RSK phosphorylated at Ser380 in cultured adherent cells, with an HRP-based colorimetric readout at 450 nm. The kit uses an indirect ELISA format: target-specific primary antibodies capture the phosphorylated protein, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

Reactivity is reported for Human, Mouse, and Rat samples, and the assay supports a dynamic range of > 5000 Cells. Because the assay is qualitative, normalisation requires two complementary approaches: a monoclonal anti-GAPDH antibody supplied as an internal positive control to normalise target absorbance values, and a Crystal Violet whole-cell staining method to determine cell density and correct for plating differences. The lysate-free, high-throughput workflow makes the kit suitable for measuring the relative amounts of phosphorylated p90 RSK in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on p90 RSK phosphorylation.

According to the associated UniProt and NCBI information, the target is the serine/threonine kinase p90RSK (gene symbol RPS6KA1; UniProt Q15418; Gene ID 6195), an AGC kinase of the RSK family that is phosphorylated and activated by Erk1 and Erk2 in response to growth factors, polypeptide hormones, and neurotransmitters. It possesses two kinase domains connected by a regulatory region and phosphorylates substrates including ribosomal protein S6, and is prominently expressed in brain structures essential for cognitive function and learning.
