p73 Transcription Factor Activity Assay

The Assay Genie p73 Transcription Factor Activity Assay quantifies the DNA-binding activity of p73, a p53-family transcription factor involved in tumor suppression, apoptosis, DNA damage response, neurodevelopment, and immune regulation. The microplate is pre-coated with a double-stranded oligonucleotide containing the p53/p73 consensus response element; activated p73 in nuclear or whole-cell lysates binds the immobilized DNA and is detected using a p73-specific primary antibody followed by an HRP-conjugated secondary antibody with TMB substrate. This approach lets researchers distinguish transcriptionally active p73 from total p73 protein levels, supporting studies of TAp73 and DeltaNp73 isoform signaling in cancer biology, neural stem cell differentiation, and stress-response pathways. Validated for human, mouse, and rat samples with cross-reactivity tested against related p53-family members.
