Description

Manufactured by Assay Genie, this colorimetric, cell-based ELISA kit is designed for the detection and quantification of phosphorylated p70 S6 Kinase at serine 418 in cell lysates. Phosphorylation at this site is known to activate the kinase's activity, making the kit useful for studying cell growth, protein synthesis, and proliferation signalling pathways. The kit is reactive with human, mouse, and rat samples and is suited to adherent cell cultures.

The assay uses an indirect ELISA format, in which a phospho-specific primary antibody captures the target and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm. It is described as lysate-free and high-throughput, and can be used to measure the relative phosphorylation status of p70 S6 Kinase in cultured cells as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators. The dynamic range is stated as greater than 5000 cells. A monoclonal antibody against human GAPDH is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining can be used to determine cell density. The associated research area is cell death, and the target gene is RPS6KB1, a member of the ribosomal S6 kinase family of serine/threonine kinases.
