Description

Designed by Assay Genie for researchers investigating cellular signalling pathways, this Cell-Based ELISA kit enables qualitative measurement of p70 S6 Kinase phosphorylation at serine 411 in cultured adherent cells. The assay uses a colorimetric detection method read at 450 nm and is reactive with Human, Mouse, and Rat samples.

Operating as a lysate-free, high-throughput indirect ELISA, the kit captures p70 S6 Kinase (Phospho-Ser411) with a phospho-specific primary antibody and detects the bound target with HRP-conjugated secondary antibodies that catalyse a colorimetric reaction upon substrate addition. It can be used both to measure relative levels of phosphorylated p70 S6 Kinase in cultured cells and to screen treatments, inhibitors, or activators for their effects on p70 S6 Kinase phosphorylation. The dynamic range is greater than 5000 cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step may be used to determine cell density.

p70 S6 Kinase (gene symbol RPS6KB1) is an AGC kinase of the RSK family that is required for cell growth and G1 cell cycle progression and is phosphorylated and activated by mTOR in mitogenic pathways downstream of PI3K. Phosphorylation at serine 411 is known to regulate the kinase's activity and function, making it a valuable marker for studying cellular signalling pathways and potential therapeutic targets in research areas including cancer, cell biology, and molecular biology.
