Description

A colorimetric cell-based ELISA kit from Assay Genie enables quantitative detection of p70 S6 Kinase phosphorylation at Ser371 in cultured cells. Designed as a lysate-free, high-throughput and sensitive assay, the kit measures relative amounts of phosphorylated p70 S6 Kinase and can be used to screen the effects of various treatments, inhibitors (e.g. siRNA or chemicals), or activators on kinase phosphorylation. Detection follows an indirect ELISA format in which p70 S6 Kinase (Phospho-Ser371) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction upon substrate addition, with absorbance measured at 450 nm.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following HRP measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalised to cell counts and adjusting for plating differences. The kit is intended for the Cell Death research area, with reactivity confirmed for Human, Mouse, and Rat samples and a dynamic range of > 5000 Cells. The target gene is RPS6KB1, classified with a Phospho sub-type designation.
