p70 S6 Kinase beta (Phospho-Ser423)Colorimetric Cell-Based ELISA Kit

The Assay Genie Cell-Based ELISA Kit enables qualitative detection of p70 S6 Kinase beta (Phospho-Ser423) phosphorylation and expression in cultured cells. It uses an indirect ELISA format in which phospho-specific or total primary antibodies capture the target, while HRP-conjugated secondary antibodies catalyse a colorimetric readout at 450 nm. The assay is reactive with human and mouse samples and has a stated dynamic range of greater than 5000 cells. It is a convenient, lysate-free, high-throughput format suitable for screening the effects of treatments, inhibitors, or activators on p70 S6 Kinase beta phosphorylation.

Normalization is supported by an included anti-GAPDH monoclonal antibody serving as an internal positive control, together with an optional Crystal Violet whole-cell staining step for cell density adjustment, allowing absorbance values to be corrected for plating variation.

p70 S6 Kinase beta (gene symbol RPS6KB2) is an AGC/RSK-family serine/threonine kinase activated by mTOR downstream of PI3K, where phosphorylation at Ser423 contributes to its activation and downstream signalling through phosphorylation of ribosomal protein S6 and translational control of 5' oligopyrimidine tract mRNAs.
