Description

Assay Genie provides a Colorimetric Cell-Based ELISA Kit built for the qualitative determination of p63 protein phosphorylation and expression profile at Ser455 in cultured cells. The kit is described as lysate-free, high throughput, and sensitive, and is recommended for use with adherent cells.

The assay uses p63 (Phospho-Ser455)-specific primary antibodies in combination with HRP-conjugated secondary antibodies; the conjugated HRP catalyses a colorimetric reaction upon substrate addition, measured at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used after the colorimetric read to determine cell density and adjust for plating variation. The kit is reactive with human samples and supports a dynamic range of > 5000 Cells.

The kit can be used to measure relative levels of phosphorylated p63 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on p63 phosphorylation. Phosphorylated p63 is a key protein in cellular signalling pathways that regulate cell growth, differentiation, and apoptosis, and aberrant phosphorylation has been linked to cancer, developmental disorders, and neurodegenerative diseases. The associated gene is TP63 (Gene ID: 8626).
