Description

From Assay Genie, this Cell-Based ELISA Kit provides a colorimetric assay for detecting and quantifying p63 protein phosphorylated at serine 395 directly in cultured cells. The kit is intended for adherent cell samples and is reactive with human, mouse, and rat specimens, with a dynamic range of > 5000 cells measured at 450 nm.

The assay uses an indirect ELISA format: phosphorylated p63 (Phospho-Ser395) is captured by a phospho-specific primary antibody, and an HRP-conjugated secondary antibody binds the Fc region of the primary to catalyse a colorimetric reaction upon substrate addition. A monoclonal antibody against human GAPDH is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step is provided to determine cell density for adjusting plating differences. The kit is lysate-free and high-throughput, making it suitable for measuring relative levels of phosphorylated p63 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on p63 phosphorylation.

Phosphorylation of p63 at serine 395 is implicated in cellular processes including proliferation, differentiation, and apoptosis, and dysregulation has been associated with cancer and developmental disorders. The kit targets the TP63 gene product (Gene ID 8626; UniProt ID Q9H3D4).
