Description
A cell-based ELISA from Assay Genie provides a qualitative, lysate-free method for detecting p62/Dok protein expression in adherent cells and for measuring p62/Dok levels in serum, plasma, and cell culture supernatants. The indirect ELISA format uses p62/Dok-specific primary antibodies for target capture, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody and catalysing a colorimetric reaction read at 450 nm.
The kit is suited to measuring relative p62/Dok levels in cultured cells and to screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators. A monoclonal antibody specific for human GAPDH is included as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density so that absorbance values can be normalised to cell amounts.
p62/Dok (DOK1; docking protein 1; Gene ID 1796, UniProt Q99704) is a scaffold protein operating in signal transduction downstream of receptor tyrosine kinases, and has been linked to cell growth, division, and survival, with dysregulation reported in cancer, metabolic disorders, and neurodegenerative conditions. Reactivity covers Human, Mouse, and Rat. The dynamic range is > 5000 Cells, with storage at 4°C for 6 months in a 96-Well Microplate format.
