Description

Designed by Assay Genie, this colorimetric cell-based ELISA kit enables researchers to detect phosphorylated p57 Kip2 (Thr310) in cultured cells. Using an indirect ELISA format, the assay provides qualitative determination of p57 Kip2 (Phospho-Thr310) levels and is suited to adherent cell samples. It shows reactivity with human, mouse, and rat samples, with a stated dynamic range of > 5000 cells and detection at 450 nm.

The lysate-free workflow is built for high-throughput applications and supports studies of cell cycle regulation, cell proliferation, and phosphorylation changes in response to treatments, inhibitors, or activators. Normalisation is addressed through two complementary approaches: a monoclonal antibody specific for human GAPDH serves as an internal positive control for the target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density. The associated gene is CDKN1C (Gene ID: 1028; UniProt ID: P49918), which encodes cyclin-dependent kinase inhibitor 1C.

Phosphorylation of p57 Kip2 at Thr310 contributes to controlling cell cycle progression and cell growth, making the assay relevant to research areas including cancer, developmental biology, and regenerative medicine. Signal generation relies on HRP-conjugated secondary antibodies binding the primary antibody Fc region, with HRP catalysing a colorimetric reaction upon substrate addition.
