Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of phosphorylated p56 Dok-2 protein at Tyr299 in cell lysates. The assay is reactive with Human and Mouse samples, uses a Colorimetric detection method at 450 nm, and has a dynamic range of > 5000 Cells. It is classified as a Phospho-specific Cell-Based ELISA within the Immunology research area.

The kit provides a convenient, lysate-free, high-throughput and sensitive format for measuring the relative amounts of phosphorylated p56 Dok-2 in cultured cells, including screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on p56 Dok-2 phosphorylation. Qualitative determination is achieved through an indirect ELISA format: p56 Dok-2 (Phospho-Tyr299) is captured by specific primary antibodies, and HRP-conjugated secondary antibodies catalyse a colorimetric reaction upon substrate addition. For normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and the Crystal Violet whole-cell staining method can be used to determine cell density. The kit is suited to adherent cell samples.

p56 Dok-2 (DOK2, Gene ID 9046, UniProt O60496) is a member of the p62dok family of proteins that interact with receptor tyrosine kinases and mediate biological responses in cell growth, differentiation, and immune response. The protein is constitutively tyrosine phosphorylated in hematopoietic progenitors isolated from chronic myelogenous leukemia (CML) patients and may be a critical substrate for p210(bcr/Abl). It contains a pleckstrin homology domain, 13 potential tyrosine phosphorylation sites, and six PXXP SH3 recognition motifs, and it binds p120 (RasGAP). The protein has a molecular weight of 45,379 Da, and phosphorylation at Tyr299 plays a crucial role in modulating its function and downstream signalling, making this kit useful for studying signal transduction pathways and potential therapeutic targets in diseases associated with dysregulated p56 Dok-2 signalling.
