Description

A colorimetric cell-based ELISA kit from Assay Genie is designed for the accurate and sensitive detection of phosphorylated p53 at serine 9 in cell lysates. Phosphorylation of p53 at serine 9 plays a critical role in regulating the activity of this tumor suppressor protein, influencing cell cycle progression, DNA repair, and apoptosis, making the kit a useful tool for cancer biology research and related fields.

The kit is a convenient, lysate-free, high-throughput, and sensitive assay that can detect p53 protein phosphorylation and expression profiles in cells. It can be used to measure the relative amounts of phosphorylated p53 in cultured cells and to screen the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on p53 phosphorylation. Qualitative determination is achieved via an indirect ELISA format in which p53 (Phospho-Ser9) is captured by phospho-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

Two normalization methods are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control, and Crystal Violet whole-cell staining can be used to determine cell density after the colorimetric measurement. The kit has human reactivity, a colorimetric 450 nm detection method, a dynamic range of > 5000 cells, and is intended for adherent cell samples.
