Description

This cell-based colorimetric ELISA kit from Assay Genie is designed for the accurate and sensitive detection of phosphorylated p53 at Ser46 in cell lysates. It provides a reliable, lysate-free, high-throughput method for studying the activation of p53 and its role in cell signalling pathways. Phosphorylation of p53 at Ser46 plays a critical role in regulating cell cycle arrest, apoptosis, and DNA repair in response to cellular stress, and the kit supports research into cancer development, progression, and treatment.

Qualitative determination of phosphorylated p53 at Ser46 is achieved by an indirect ELISA format. Target-specific primary antibodies capture p53 (Phospho-Ser46), while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyzes a colorimetric reaction upon substrate addition, measured at 450 nm, with a dynamic range of > 5000 Cells. Reactivity is human. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing absorbance values. After HRP measurement, the Crystal Violet whole-cell staining method can be used to determine cell density, enabling results to be normalized to cell amounts and account for plating differences.

The assay is intended for use with adherent cells and for measuring the relative amounts of phosphorylated p53 in cultured cells, including screening the effects of treatments, inhibitors such as siRNA or chemicals, and activators on p53 phosphorylation. Database information for the target includes Gene ID: 7157, UniProt ID: P04637, and Gene Symbol: TP53. The sub-type is Phospho.
