The p53 (Phospho-Ser33) Transcription Factor Activity Assay from Assay Genie is a non-radioactive DNA-binding immunoassay that measures the DNA-binding activity of p53 when phosphorylated at serine 33. The microplate is pre-coated with a double-stranded oligonucleotide containing a consensus p53 response element, capturing activated phospho-p53 from nuclear and cell lysates. A phospho-Ser33-specific primary antibody is then applied, and colorimetric readout enables quantification relative to a standard or reference sample.

This ELISA-based assay supports studies of the DNA damage response, apoptosis signaling, and cell cycle checkpoint regulation across cancer biology, toxicology, and chemotherapeutic response research. The kit has been validated for human, mouse, and rat samples, supporting cross-species comparisons in translational research workflows. A recommended sample volume of 10 uL nuclear extract per well is used, and lysates should be stored at -80 °C and protected from repeated freeze-thaw cycles. All required reagents, wash buffers, detection antibodies, and TMB substrate are supplied to complete the 96 Assays workflow without radioactive probes.
