The p53 (Phospho-Ser20) Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA kit that quantifies the DNA-binding activity of p53 phosphorylated at serine 20, a modification catalyzed primarily by ATM kinase in response to DNA double-strand breaks. This phospho-specific activity readout distinguishes activated, DNA-damage-responsive p53 from total p53 protein levels, providing a functional measure of tumor suppressor engagement with its consensus response elements in promoter regions.

Researchers studying apoptosis, genotoxic stress, oncogene activation, and chemotherapeutic response apply this assay on nuclear and cell lysates from human, mouse, and rat samples. The microplate format supports high-throughput screening of compound libraries or time-course experiments following irradiation, etoposide, or other DNA-damaging agent exposure. Each well is coated with oligonucleotides containing the p53 consensus binding sequence, capturing only the functionally active phospho-Ser20 p53 fraction from your lysate. Store unopened components frozen at -20 degrees C and avoid repeated freeze-thaw cycles to preserve antibody activity.
