Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the qualitative detection of phosphorylated p47 phox at Ser359 in cultured human cells. The assay uses a colorimetric readout at 450 nm in an indirect ELISA format, in which p47 phox (Phospho-Ser359) is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies. It is described as lysate-free and suitable for high-throughput screening of treatments, inhibitors, or activators that affect p47 phox phosphorylation. The stated dynamic range is > 5000 cells and the kit is intended for use with adherent cells.

Two normalisation approaches are included: an anti-GAPDH monoclonal antibody that serves as an internal positive control, and Crystal Violet whole-cell staining to determine cell density and adjust for plating differences. The target, p47phox, is the 47-kilodalton cytosolic subunit of the NADPH oxidase complex (gene symbol NCF1, UniProt ID P14598) and contains 2 SH2 domains. Phosphorylation at Ser359 is reported as a key regulatory event in NADPH oxidase activation, and the kit is presented as relevant to research on oxidative stress, immune cell activation, inflammation, cardiovascular disorders, neurodegenerative diseases, and chronic granulomatous disease.
