Description

A sensitive, high-throughput cell-based assay from Assay Genie designed to detect p47 phox protein phosphorylation and expression directly in cultured cells without a lysate preparation step. The kit uses an indirect ELISA format in which target-specific primary antibodies capture p47 phox (Phospho-Ser304) and HRP-conjugated secondary antibodies catalyze a colorimetric reaction read at 450 nm. It is reactive with human samples and validated for adherent cells, with a quoted dynamic range of > 5000 cells.

P47-Phox is a 47 kDa cytosolic subunit of the NADPH oxidase complex that plays a key role in the production of reactive oxygen species (ROS) in immune cells. Phosphorylation at Ser304 is essential for its activation and function in ROS production, linking this readout to immune responses and inflammatory processes. By measuring levels of phosphorylated P47-Phox, the kit supports research into ROS regulation in disease states such as inflammation, cancer, and autoimmune disorders, and can be used to screen the effects of treatments, inhibitors (including siRNA or chemicals), or activators on p47 phox phosphorylation. The associated gene is NCF1, and mutations in this and other NADPH oxidase subunits are linked to chronic granulomatous disease.

A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization of target absorbance values, and Crystal Violet whole-cell staining is provided to determine cell density for further data normalization.
