Description

From Assay Genie, this Cell-Based ELISA Kit is designed to detect and quantify levels of phosphorylated p44/42 MAP kinase, also known as ERK1/2, in cell lysates. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, offering a lysate-free, high-throughput workflow for measuring relative phosphorylation of p44/42 MAP Kinase in cultured cells. It supports screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on p44/42 MAP Kinase phosphorylation.

Reactivity is reported for Human, Mouse, and Rat, with a dynamic range of > 5000 Cells. The workflow captures the target with phospho-specific primary antibodies, while HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining is recommended to determine cell density so absorbance values can be normalized to cell amounts.

p44/42 MAP kinase (gene symbol MAPK1/MAPK3; UniProt P27361/P28482) is a serine/threonine kinase central to MAPK signaling cascades that regulate cell growth, proliferation, and differentiation. Phosphorylation at Tyr204 is a key regulatory event in its activation, making this phospho-specific assay a useful tool for studying signaling pathways and potential therapeutic targets. The target is classified as a phospho-subtype with EC number 2.7.11.24.
