Description

Designed for the sensitive and accurate detection of phosphorylated p38 MAPK at tyrosine 322 in cultured cells, this colorimetric cell-based ELISA from Assay Genie provides a lysate-free, high-throughput workflow. The kit uses an indirect ELISA format in which target-specific primary antibodies are combined with HRP-conjugated secondary antibodies; upon substrate addition, a colorimetric reaction is measured at 450 nm. It can be used to measure relative levels of phosphorylated p38 MAPK and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on p38 MAPK phosphorylation.

Reactivity is reported for human, mouse, and rat samples, with a dynamic range of > 5000 cells. Because the assay is qualitative, two normalization strategies are provided: a monoclonal GAPDH antibody serves as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust the absorbance values to plating differences.

p38 MAPK is a key signaling molecule involved in cellular responses to stress, inflammation, and apoptosis, and phosphorylation at tyrosine 322 is essential for its activation and regulation of downstream signaling pathways. Dysregulation of p38 MAPK has been implicated in cancer, inflammatory disorders, and neurodegenerative diseases, making this kit a useful tool for studying p38 MAPK signaling and identifying potential therapeutic targets.
